I found that ths trophie can be glitched and no full second playthrough is needed. On missing this trophie on my first play through, I started my second playthrough upon selecting resume game I started from the start played for a few minutes, left the game then resumed the game at the very last boss and got the trophie after beating him again.

I tried this a couple of ways and can't get the trophy to ding, I think you where just lucky.
