You never need to get the DLC trophies in any game to get the platinum. And you can't get any DLC trophies with the free map "Synthesisia".

the free map can help you with time trials though. any stars you get on that map will go towards the total 50 that you need for the trophy, so it can make it slightly easier for you.

Well only the time trials will be easier. The speedruns will still be extremely difficult.
